ZLWH-67 is a β-Carboline derivative and Antibacterial agent. ZLWH-67 inhibits DNA synthesis, suppresses biofilm formation, and increases reactive oxygen species (ROS) levels. ZLWH-67 exhibits potent in vitro antibacterial activity against MRSA (MIC = 0.5-4 μg/mL), S. epidermidis (MIC = 4 μg/mL), E. faecalis (MIC = 4-8 μg/mL), and S. pneumoniae (MIC = 16 μg/mL). ZLWH-67 displays anti-MRSA effects in murine skin and pneumonia infection models[1].

Animal Model: BALB/c (female, 6−8 weeks old)[1]
Dosage: 0.4 mg/kg
Administration: topical; twice daily; 4 days
Result: Reduced bacterial burden in skin tissues (log10 CFU/0.1 g tissue decreased relative to controls, P=0.0364).
Promoted wound closure relative to controls.
Significantly reduced levels of proinflammatory cytokines TNF-α, IL-6, and IL-1β in skin tissues (P=0.0009, P=0.0239, P=0.0201 respectively).
Alleviated skin tissue pathology including necrosis, inflammatory exudate, and hemorrhage.
Animal Model: C57BL/6 (female, 6−8 weeks old)[1]
Dosage: 100 mg/kg/day
Administration: daily
Result: In the lethal pneumonia model, significantly improved mouse survival rate relative to controls.
Alleviated infection-related clinical symptoms and promoted body weight recovery relative to controls.
In the sublethal pneumonia model, reduced bacterial burden in lung tissues (log10 CFU/mL tissue decreased relative to controls, P=0.0047).
Significantly reduced levels of proinflammatory cytokines TNF-α, IL-6, and IL-1β in lung tissues (P=0.0008, P=0.0016, P<0.0001 respectively).
Alleviated lung tissue pathology including alveolar wall thickening, epithelial desquamation, inflammatory infiltration, and hemorrhage.
