KS-99 is a dual inhibitor with both tubulin polymerization inhibitory activity and Akt pathway inhibitory activity. KS-99 inhibits cancer cell proliferation and induces cancer cell apoptosis. KS-99 can be used in research related to colorectal cancer, breast cancer, lung epithelial cancer and melanoma[1].

Cell Cytotoxicity Assay[1]

Cell Line: human colorectal cancer HT29 cells, human breast cancer MCF-7 cells, human lung epithelial carcinoma A549 cells, human melanoma UACC903 cells, mouse embryonic fibroblasts (MEF) cells
Concentration: 0.5-50 μM
Incubation Time: 48 h
Result: Exhibited an IC50 of 1.14 μM against HT29 cells, 24.70 μM against MCF-7 cells, 2.53 μM against A549 cells, 2.23 μM against UACC903 cells, and an IC50 > 20 μM against MEF cells.

Apoptosis Analysis[1]

Cell Line: human colorectal cancer HT29 cells
Concentration: 1.25 μM, 2.5 μM, 5.0 μM
Incubation Time: 24 h
Result: Induced apoptosis in 11.47% of HT29 cells at 1.25 μM, 39.46% at 2.5 μM, and 66.98% at 5.0 μM.

Apoptosis Analysis[1]

Concentration: 5 μM
Incubation Time: 48 h
Result: Resulted in a significant number of TUNEL-positive HT29 cells, indicating DNA fragmentation as a hallmark of apoptosis.

Western Blot Analysis[1]

Incubation Time: 24 h
Result: Reduced Akt protein levels and strongly suppressed the phosphorylation of Akt, eliminating detectable phosphorylated Akt at 1 μM and 2 μM.
