生物活性

Misitatug blivedotin is an antibody-drug conjugate (ADC) targeting mesothelin (MSLN). Misitatug blivedotin binds to mesothelin and exhibits dose‐dependent antitumor activity. Misitatug blivedotin can be used for the research of ovarian cancer, non-squamous non-small-cell lung carcinoma, cervical cancer[1][2].

Misitatug blivedotin (0.7-3 mg/kg；静脉注射；每周一次，给药 1 或 3 次) 在皮下植入 300 万 OVCAR3‐MSLN‐3# 或 H292‐MSLN‐9C8 细胞的雌性 Balb/c 裸鼠体内表现出剂量依赖性的抗肿瘤活性[2]。

Animal Model: Balb/c nude mice (female; ages 4–5 weeks; implanted subcutaneously with 3 million OVCAR3‐MSLN‐3# or H292‐MSLN‐9C8 cells for a xenograft model)[3]
Dosage: 0.7 mg/kg; 0.75 mg/kg; 1.5 mg/kg; 2.1 mg/kg; 3 mg/kg
Administration: i.v.; Q1W × 1 or Q1W × 3
Result: Had moderate activity at 0.75 mg/kg with single dosing, but tumor volume was less than the initial volume of tumors treated at 2.1 mg/kg until 11 days after dosing.
Significantly reduced the tumors 7 days after the first dosing at 1.5 and 3 mg/kg with multiple dosing.
Fully regressed tumors at 3 mg/kg for 10 days and at 1.5 mg/kg for 17 days, but the tumor continued to grow after three administrations at 0.75 mg/kg.
