NFYi5 is a nuclear transcription factor-Y (NF-Y) inhibitor. NFYi5 disrupts the binding of NF-Y to DNA, accelerates the ubiquitin-independent degradation of the NF-YA subunit, and reduces the transcriptional activity of NF-Y. As an antimitotic agent, NFYi5 decreases the mRNA levels of NF-Y target genes without affecting the expression of housekeeping genes, and inhibits cell proliferation. NFYi5 can be used in the research of tissue fibrosis[1].

NFYi5 (0.01-100 μM; 18 h) 可剂量依赖性地抑制 NF-Y 依赖性报告基因活性，在人心肌成纤维细胞中的 IC50 为 19.95 μM，在大鼠心肌成纤维细胞中的 IC50 为 12.73 μM，同时不影响非 NF-Y 启动子活性[1]。
NFYi5 (10-20 μM; 18 h) 可在人源和大鼠心肌成纤维细胞中选择性降低 NF-Y 靶基因的 mRNA 水平，且不影响管家基因的表达[1]。
NFYi5 (5-20 μM; 24-48 h) 可剂量依赖性地抑制人和大鼠心脏成纤维细胞的增殖[1]。

Incubation Time: 18 h
Result: Reduced mRNA levels of NF-Y target genes CCNA2 and CCNB1 in human cardiac fibroblasts without altering GAPDH levels.
Dose-dependently reduced mRNA levels of NF-Y target genes CCNA2, CCNB1, ECT2, and DDX11 in rat cardiac fibroblasts, while leaving housekeeping gene mRNA levels (TBP, 36B4, GAPDH, UBC) unchanged.
