ON203 is a fully human IgG1 monoclonal antibody targeting oxidized macrophage migration inhibitory factor (oxMIF), with an EC50 of 0.3 nM. ON203 selectively binds to oxMIF, enhances binding to FcγRIIIA and FcγRIIA, and triggers antibody-dependent cellular cytotoxicity (ADCC) and antibody-dependent cellular phagocytosis (ADCP). ON203 induces the release of TNFα, IFNγ, MCP-1, and IL6, inhibits tumor growth, reduces the proliferative capacity of tumor cells, and decreases tumor angiogenesis. ON203 can be used in the research of prostate cancer[1].

Animal Model: NMRI nude (male, 6-9 weeks old, subcutaneous injection of 5×106 PC3 human prostate adenocarcinoma cells)[1]
Dosage: 5 mg/kg; 20 mg/kg
Administration: i.p.; every other day; 15 times
Result: Significantly inhibited tumor growth with durable response during post-treatment washout period at 20 mg/kg.
Showed no meaningful difference from controls at 5 mg/kg.
Reduced Ki67-positive cell count, tumor vascularization, and tumor cell intravasation into blood vessels at 20 mg/kg.
Maintained stable body weight across all groups with no acute toxicity observed.
