MCE收集了近千个临床阶段已披露的变构调节剂，分析 PDB 数据库中解析的变构相关蛋白 - 配体复合物结构，提取其核心药效团与优势骨架，采用 “骨架衍生 + 变构理化规则过滤” 设计策略，基于通用变构口袋特征，严格遵循变构结合最优理化原则对衍生化合物进行二次筛选：分子量300–500 Da、氢键供体（HBD ≤ 3）、氢键受体（HBA = 3–8）、适中极性表面积（PSA = 70–120 Ų）、可旋转键（≤ 6）、高刚性骨架、适中疏水性（cLogP = 1.0–3.8）、无强电离基团，具备高刚性、形状互补性，适配结合浅型、动态、疏水主导的变构口袋。

本库包含4315 个结构多样、类药性质优异的化合物，专为变构药物研发、变构位点靶向及变构调节剂筛选设计，适用于激酶、GPCR等多种靶点。库内化合物为相似度>0.6的临床阶段的变构调节剂类似物，兼具成药性与变构结合潜力，是变构药物早期发现的高效工具。
