SIBP-03 is a specifical anti-HER3 antibody. SIBP-03 binds strongly and specifically to recombinant human HER3 protein. SIBP-03 inhibits HER3 activation, as well as the downstream PI3K/AKT signaling pathway. SIBP-03 exhibits anticancer activity against squamous cell carcinoma, non-small cell lung cancer, gastric cancer, and breast cancer. SIBP-03 synergistically enhances the antitumor activity of DS-8201 (HY-138298A) and Cetuximab (HY-P9905)[1][2].

(In Vitro)

SIBP-03 (10-4-104 ng/mL) 可与重组人 HER3 蛋白发生强特异性结合，而与重组人 EGFR、HER2 或 HER4 蛋白未检测到结合[1]。
SIBP-03 结合过表达 HER3 的 MC38/HER3 细胞表面的 HER3，而与 HER3 阴性的 MC38 细胞无可检测到的结合[1]。
SIBP-03 (0.01-10 μg/mL；FaDu/BT-474 处理 24 h，NRG1 刺激细胞预孵育 2 h) 有效抑制 FaDu、BT-474 以及 NRG1 刺激的 MCF7 和 BT-474 细胞中配体非依赖性和配体依赖性的 HER3 激活，以及下游的 PI3K/AKT 信号通路[1]。
SIBP-03 (0.01-100 μg/mL; 120 h) 在 FaDu、A549、BT-474、SK-BR-3 和 NCI-N87 细胞中仅发挥微弱的抗增殖作用，但可强效抑制 NRG1 刺激的 MCF7 细胞增殖，100 μg/mL 处理后抑制率达 74.8%[1]。
SIBP-03 (10-100 μg/mL; 1 h) 在 10 和 100 μg/mL 浓度下不会在 FaDu 细胞或过表达 HER3 的 MC38/HER3 细胞中诱导补体依赖的细胞毒性[1]。
SIBP-03 (10-100 μg/mL; 5 h) 可在过表达 HER3 的 MC38/HER3 细胞中诱导 ADCC 活性，但在 10 和 100 μg/mL 浓度下无法在 BT-474 细胞中诱导该活性[1]。
SIBP-03 (10-100 μg/mL; 5 h) 在 10 μg/mL 和 100 μg/mL 浓度下，可在人乳腺癌细胞 BT-474 以及过表达 HER3 的 MC38/HER3 细胞中诱导抗体依赖性细胞吞噬活性[1]。
SIBP-03 (30 μg/mL; 144 h) 可协同增强 DS-8201 在 SK-BR-3 和 MDA-MB-453 细胞中的抗增殖活性[2]。

Result: Exhibited weak anti-proliferative effects, with inhibition rates ranging from 10.0% to 32.9% at 100 μg/mL in FaDu, A549, BT-474, SK-BR-3, and NCI-N87 cells.
Induced strong anti-proliferative effects, with 74.8% inhibition at 100 μg/mL in NRG1-stimulated MCF7 cells.

SIBP-03 (0.2-2 mg/kg；静脉注射) 在 FaDu 异种移植小鼠模型中展现出强效抗肿瘤活性，在 2 mg/kg 剂量下的 TGI 达 116.3%，且单次静脉给药后可对 HER3/AKT 信号通路产生长达 168 h 的持续性抑制[1]。
SIBP-03 (6 mg/kg；静脉注射) 在 A549 异种移植小鼠模型中表现出显著的抗肿瘤活性，6 mg/kg 剂量下的肿瘤生长抑制率 (TGI) 为 57.5%[1]。
SIBP-03 (20 mg/kg；静脉注射) 在 NCI-N87 异种移植小鼠模型中表现出显著的抗肿瘤活性，20 mg/kg 剂量下的肿瘤生长抑制率 (TGI) 为 50.8%[1]。
SIBP-03 (0.6 mg/kg；静脉注射) 在 FaDu 异种移植瘤小鼠中单处理实现了 96% 的 TGI，且与 Cetuximab (HY-P9905) 联用可诱导 152% 的 TGI，同时使 66.7% 的治疗小鼠出现肿瘤消退[1]。
