SMN2 modulator-1 is a brain-penetrant survival motor neuron (SMN) modulator. SMN2 modulator-1 post-translationally stabilizes SMN protein and increases SMN protein levels independent of SMN2 transcription. SMN2 modulator-1 can be used for the research of spinal muscular atrophy[1].

体外研究
(In Vitro)

SMN2 modulator-1 (Compound 27) (0.29-30 μM；24 小时) 可强效激活 SMN2 报告细胞中的 SMN2 表达，其 EC50 为 0.29 μM，活性可达对照组的 190%[1]。
SMN2 modulator-1 (0.4-10 μM；48 小时) 可呈剂量依赖性地使 GM003813T 型 SMA 人源成纤维细胞中的 SMN 蛋白水平升高；在 2 μM 处理 48 小时的条件下，蛋白水平约升高 2 倍[1]。
SMN2 modulator-1 在 pH 7.4 的条件下，动力学溶解度为 31 μM，小鼠肝微粒体半衰期为 39 分钟，人肝微粒体半衰期大于 120 分钟，小鼠血浆半衰期为 326 分钟[1]。
SMN2 modulator-1 (1-30 μM；30 分钟-8 小时) 不会抑制 HEK293 细胞中的蛋白酶体或自噬，也不会在体外抑制 HPV-16 E6 介导的 p53 降解，表明其 SMN 稳定效应并非源于全局蛋白质降解抑制[1]。
SMN2 modulator-1 在 Caco-2 细胞模型中表现出良好的肠道通透性，其顶侧到底侧的 Papp 值为 15 × 10-6 cm/s[1]。
