Uniblue A sodium is a reactive protein stain that can be used in the covalent pre-gel staining of the protein (Ex=594 nM)[1].

1. 准备衍生化缓冲液：100 mM 碳酸氢钠，10% SDS，pH 值为 8-9 。
2. 向 90 μL 蛋白质溶液中加入 10 μL 200 mM 的 Uniblue A 溶液 (该溶液溶解于衍生化缓冲液中)。
3. 将样本在 100°C 下加热 1 分钟以进行染色。
4. 加入 100 μL 由 10% 甘油和 20 mM 二硫苏糖醇（DTT）组成的还原溶液，该溶液置于 200 mM Tris 缓冲液中，缓冲液的 pH 值为 6.8。
5. 过量的 Uniblue A 与 Tris 反应，生成一种蓝色化合物，该化合物在电泳过程中可作为运行前的指示物。
6. 该样品在 100°C 的温度下再加热一分钟，以实现高效的还原过程，并随后自然冷却至室温。
7. 加入 20 μL 含 550 mM 碘乙酰胺 (IAA) 的烷基化溶液，然后室温下孵育 5 分钟。

作为阳性对照，衍生化缓冲液中加入了牛血清白蛋白 (BSA)，其浓度为 10 mg/mL。
