相比传统正构位点的高度保守，变构位点具有低保守性、强疏水性、弱极性、空间受限且动态隐式的结构特征，其核心结构与正构口袋存在显著差异——变构口袋多为蛋白构象变化形成的动态沟槽、亚基界面裂缝或浅型凹陷，而非正构位点的刚性“锁孔”结构，空间约束更宽松，因此具备高选择性、低脱靶风险的优势，已成为新药研发的重要方向。

MCE基于变构口袋的动态性、疏水性及狭长型空间特征，对片段进行定向修饰与筛选。筛选标准严格贴合变构结合需求：分子量控制在120–280 Da（适配片段库小分子、高衍生性的核心需求）、氢键供体（HBD ≤ 2、氢键受体（HBA ≤ 3）、极性表面积（PSA = 30–80 Ų）、可旋转键（≤ 2）、适中疏水性（cLogP = 1–3.5），无强电离基团，兼具适度刚性与构象柔性，适配口袋的动态变化。同时结合主转动惯量（PMI）分析结果，得到高3D多样性的片段，该类片段与变构口袋的形状互补性较好，保证片段能顺利进入变构口袋并形成稳定结合，同时后续优化的衍生空间。

本库包含1800个结构多样、类药性质优良的片段分子，适配变构药物研发、针对变构位点设计优化，兼具变构结合潜力和“结构简洁、衍生性强”的特点，有效提升变构筛选/衍生的hit率，为变构药物早期研发提供高效、精准的筛选工具。
