生物活性

Octanoylcarnitine is an orally active medium-chain acylcarnitine transport intermediate in fatty acid β-oxidation. Octanoylcarnitine is converted to octanoyl-CoA by carnitine acetyltransferase (CrAT), which then generates energy via β-oxidation in mitochondria of the heart and skeletal muscle. Octanoylcarnitine enhances grip strength and treadmill endurance, alleviates lactic acidosis, distributes in muscle and heart tissues, increases free carnitine levels, and mitigates mitochondrial stress. Octanoylcarnitine is associated with long-chain fatty acid metabolism, shows a positive correlation with subcutaneous fat area in patients with metastatic pancreatic ductal adenocarcinoma, and is closely related to central retinal artery occlusion (CRAO)[1][2][3].

Animal Model: Long-Chain Acyl-CoA Dehydrogenase Knockout (LCAD-KO) (male, 12-16 weeks, LC-FAOD model)[1]
Dosage: 0.5 mg/g (exercise testing); 3% w/w (chronic feeding)
Administration: i.g.; single dose, 20 minutes prior to exercise testing; i.g.; mixed into powdered diet, ad libitum for 10 days
Result: Significantly increased treadmill running distance compared to vehicle control.
Significantly reduced post-run blood lactate levels compared to vehicle control.
Did not alter serum levels of diagnostic long-chain acylcarnitine C14:1-carnitine.
Increased serum levels of downstream short-chain acylcarnitines (C8, C6, C4, C2, free carnitine) to normal or above-normal levels.
Significantly increased free carnitine levels in liver, heart, and muscle.
Significantly increased octanoylcarnitine levels in liver and muscle.
Animal Model: muscle-specific Carnitine Palmitoyltransferase 2 Knockout (CPT2ᵐᴷᴼ) (male, LC-FAOD model)[1]
Dosage: 0.5 mg/g
Administration: i.g.; single dose, 20 minutes prior to functional testing
Result: Significantly increased treadmill top speed compared to vehicle control.
Significantly increased grip strength compared to vehicle control.
